Bad Phospho-Regulation
Background: Bad is a member of the BCL-2 family of regulators involved in programmed cell death. This protein positively regulatescell apoptosis by forming heterodimers with BCL-xL and BCL-2, and reversing their death repressor activity. Proapoptoticactivity of this protein is regulated through its phosphorylation. Protein kinases AKT IKK, and MAP kinases, as well asprotein phosphatase calcineurin are found to be involved in the regulation of this Bad activity. Phosphorylation of Badoccurs on one or more of Ser-26, Ser-112, Ser-136, and Ser-155 in response to survival stimuli, which blocks its pro-apoptotic activity. Phosphorylation on Ser-136 or Ser-112 promotes heterodimerization with 14-3-3 proteins. This interaction then facilitates the phosphorylation at Ser-155, a site within the BH3 motif, leading to the release of Bcl-xL andthe promotion of cell survival. Ser-26 is phosphorylated by IKK leading to phosphorylation of C-terminal serine sites and disruption of binding to Bcl-xL. This inactivation of Bad inhibits TNFalpha-induced apoptosis independent of NF-kappaB activity.

Product Notes
The Human, Rat, Mouse Bad (Catalog #AAA474196) is an Antibody Sampler Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA474196 Antibody Sampler Kit recognizes Human, Rat, Mouse Bad. AAA Biotech's Bad Phospho-Regulation can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), ELISA (EIA), Immunocytochemistry (ICC), Immunohistochemistry (IHC). Researchers should empirically determine the suitability of the Bad for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Bad Phospho-Regulation, Antibody Sampler Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
