Intended Uses: This ZP3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human ZP3. This ELISA kit for research use only!

Principle of the Assay: ZP3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-ZP3 antibody and an ZP3-HRP conjugate. The assay sample and buffer are incubated together with ZP3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ZP3 concentration since ZP3 from samples and ZP3-HRP conjugate compete for the anti-ZP3 antibody binding site. Since the number of sites is limited, as more sites are occupied by ZP3 from the sample, fewer sites are left to bind ZP3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ZP3 concentration in each sample is interpolated from this standard curve.

NCBI Description

The zona pellucida is an extracellular matrix that surrounds the oocyte and early embryo. It is composed primarily of three or four glycoproteins with various functions during fertilization and preimplantation development. The protein encoded by this gene is a structural component of the zona pellucida and functions in primary binding and induction of the sperm acrosome reaction. The nascent protein contains a N-terminal signal peptide sequence, a conserved ZP domain, a C-terminal consensus furin cleavage site, and a transmembrane domain. It is hypothesized that furin cleavage results in release of the mature protein from the plasma membrane for subsequent incorporation into the zona pellucida matrix. However, the requirement for furin cleavage in this process remains controversial based on mouse studies. A variation in the last exon of this gene has previously served as the basis for an additional ZP3 locus; however, sequence and literature review reveals that there is only one full-length ZP3 locus in the human genome. Another locus encoding a bipartite transcript designated POMZP3 contains a duplication of the last four exons of ZP3, including the above described variation, and maps closely to this gene.
