Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human WT1 antibody. WT1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human WT1 Antibody is added and binds to WT1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated WT1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human WT1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

The Wilms tumor suppressor gene WT1 appears to play a role in both transcriptional and posttranscriptional regulation of certain cellular genes. This gene encodes a WT1-associating protein, which is a ubiquitously expressed nuclear protein. Like WT1 protein, this protein is localized throughout the nucleoplasm as well as in speckles and partially colocalizes with splicing factors. Alternative splicing of this gene results in several transcript variants encoding three different isoforms. [provided by RefSeq, Jul 2012]
