Intended Uses: This ATP6V0A1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human ATP6V0A1. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: ATP6V0A1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ATP6V0A1 antibody and an ATP6V0A1-HRP conjugate. The assay sample and buffer are incubated together with ATP6V0A1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ATP6V0A1 concentration since ATP6V0A1 from samples and ATP6V0A1-HRP conjugate compete for the anti-ATP6V0A1 antibody binding site. Since the number of sites is limited, as more sites are occupied by ATP6V0A1 from the sample, fewer sites are left to bind ATP6V0A1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ATP6V0A1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a component of vacuolar ATPase (V-ATPase), a multisubunit enzyme that mediates acidification of eukaryotic intracellular organelles. V-ATPase dependent organelle acidification is necessary for such intracellular processes as protein sorting, zymogen activation, receptor-mediated endocytosis, and synaptic vesicle proton gradient generation. V-ATPase is composed of a cytosolic V1 domain and a transmembrane V0 domain. The V1 domain consists of three A and three B subunits, two G subunits plus the C, D, E, F, and H subunits. The V1 domain contains the ATP catalytic site. The V0 domain consists of five different subunits: a, c, c', c", and d. Additional isoforms of many of the V1 and V0 subunit proteins are encoded by multiple genes or alternatively spliced transcript variants. This gene encodes one of three A subunit proteins and the encoded protein is associated with clathrin-coated vesicles. Three transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
