The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Tyrosine 3/Tryptophan 5 Monooxygenase Activation Protein Epsilon (YWHAe). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Tyrosine 3/Tryptophan 5 Monooxygenase Activation Protein Epsilon (YWHAe). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Tyrosine 3/Tryptophan 5 Monooxygenase Activation Protein Epsilon (YWHAe), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Tyrosine 3/Tryptophan 5 Monooxygenase Activation Protein Epsilon (YWHAe) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

This gene product belongs to the 14-3-3 family of proteins which mediate signal transduction by binding to phosphoserine-containing proteins. This highly conserved protein family is found in both plants and mammals, and this protein is 100% identical to the mouse ortholog. It interacts with CDC25 phosphatases, RAF1 and IRS1 proteins, suggesting its role in diverse biochemical activities related to signal transduction, such as cell division and regulation of insulin sensitivity. It has also been implicated in the pathogenesis of small cell lung cancer. Two transcript variants, one protein-coding and the other non-protein-coding, have been found for this gene.
