Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for SPI1 elisa kit
Description: This Human Transcription factor PU.1 (SPI1) ELISA Kit employs a two-site sandwich ELISA to quantitate SPI1 in samples. An antibody specific for SPI1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SPI1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SPI1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SPI1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Transcription factor PU.1 (SPI1) is an ETS-domain transcription factor that activates gene expression during myeloid and B-lymphoid cell development. The nuclear protein binds to a purine-rich sequence known as the PU-box found near the promoters of target genes, and regulates their expression in coordination with other transcription factors and cofactors. SPI1 can also regulate alternative splicing of target genes. Multiple transcript variants encoding different isoforms have been found for SPI1.

NCBI Description

This gene encodes an ETS-domain transcription factor that activates gene expression during myeloid and B-lymphoid cell development. The nuclear protein binds to a purine-rich sequence known as the PU-box found near the promoters of target genes, and regulates their expression in coordination with other transcription factors and cofactors. The protein can also regulate alternative splicing of target genes. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

Uniprot Description

PU.1: Binds to the PU-box, a purine-rich DNA sequence (5'- GAGGAA-3') that can act as a lymphoid-specific enhancer. This protein is a transcriptional activator that may be specifically involved in the differentiation or activation of macrophages or B- cells. Also binds RNA and may modulate pre-mRNA splicing. Binds DNA as a monomer. Interacts with CEBPD and NONO. Interacts with RUNX1 and SPIB. Interacts with GFI1; the interaction represses SPI1 transcriptional activity. Highly expressed in both FV-P and FV-A-induced erythro- leukemia cell lines that have undergone rearrangements of the Spi- 1 gene due to the insertion of SFFV. Belongs to the ETS family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Human SPI1 spi1 (Catalog #AAA9714192) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9714192 ELISA Kit recognizes Human SPI1. It is sometimes possible for the material contained within the vial of "Transcription Factor PU.1 (SPI1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
