Intended Uses: This sandwich kit is for the accurate quantitative detection of Human T cell immunoreceptor with Ig and ITIM domains (also known as TIGIT) in serum, plasma, cell culture supernates, Ascites, tissue homogenates or other biological fluids.

Principle of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Human TIGIT antibody. TIGIT present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human TIGIT Antibody is added and binds to TIGIT in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated TIGIT antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human TIGIT. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

NCBI Description

This gene encodes a member of the PVR (poliovirus receptor) family of immunoglobin proteins. The product of this gene is expressed on several classes of T cells including follicular B helper T cells (TFH). The protein has been shown to bind PVR with high affinity; this binding is thought to assist interactions between TFH and dendritic cells to regulate T cell dependent B cell responses.[provided by RefSeq, Sep 2009]

Binds with high affinity to the poliovirus receptor (PVR) which causes increased secretion of IL10 and decreased secretion of IL12B and suppresses T-cell activation by promoting the generation of mature immunoregulatory dendritic cells.
