Intended Uses: This SUMO2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human SUMO2. This ELISA kit for research use only!

Principle of the Assay: SUMO2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-SUMO2 antibody and an SUMO2-HRP conjugate. The assay sample and buffer are incubated together with SUMO2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SUMO2 concentration since SUMO2 from samples and SUMO2-HRP conjugate compete for the anti-SUMO2 antibody binding site. Since the number of sites is limited, as more sites are occupied by SUMO2 from the sample, fewer sites are left to bind SUMO2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SUMO2 concentration in each sample is interpolated from this standard curve.

SUMO2: Ubiquitin-like protein that can be covalently attached to proteins as a monomer or as a lysine-linked polymer. Covalent attachment via an isopeptide bond to its substrates requires prior activation by the E1 complex SAE1-SAE2 and linkage to the E2 enzyme UBE2I, and can be promoted by an E3 ligase such as PIAS1-4, RANBP2 or CBX4. This post-translational modification on lysine residues of proteins plays a crucial role in a number of cellular processes such as nuclear transport, DNA replication and repair, mitosis and signal transduction. Polymeric SUMO2 chains are also susceptible to polyubiquitination which functions as a signal for proteasomal degradation of modified proteins. Homotrimer (Potential). Crystal packing analysis suggests a possible trimeric assembly, of which the biological significance remains to be determined. Interacts with SAE2 and UBE2I.
