* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Background: The highest amino acid sequence similarity found was with the myeloid-specific CD33 molecule, and similar to other sialoadhesin molecules, AIRM1 appears to mediate sialic acid-dependent ligand recognition. By analyzing genomic DNA samples derived from a panel of human/hamster somatic cell hybrids, Falco et al. (1999) mapped the SIGLEC7 gene to chromosome 19. They noted that other members of the sialoadhesin family map to chromosome 19, including CD33, SIGLEC6, SIGLEC5, myelin-associated glycoprotein (MAG), and CD22. Therefore, SIGLEC7 is probably located on 19q13.3, where these other family members have been localized. Human tissues show strong expression of Siglec-7 mRNA in spleen, peripheral blood leukocytes, and liver.

SIGLEC7: Putative adhesion molecule that mediates sialic-acid dependent binding to cells. Preferentially binds to alpha-2,3- and alpha-2,6-linked sialic acid. Also binds disialogangliosides (disialogalactosyl globoside, disialyl lactotetraosylceramide and disialyl GalNAc lactotetraoslylceramide). The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface. In the immune response, may act as an inhibitory receptor upon ligand induced tyrosine phosphorylation by recruiting cytoplasmic phosphatase(s) via their SH2 domain(s) that block signal transduction through dephosphorylation of signaling molecules. Mediates inhibition of natural killer cells cytotoxicity. May play a role in hemopoiesis. Inhibits differentiation of CD34+ cell precursors towards myelomonocytic cell lineage and proliferation of leukemic myeloid cells (in vitro). Belongs to the immunoglobulin superfamily.
