Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Proline/serine-rich coiled-coil protein 1 (PSRC1) ELISA Kit employs a two-site sandwich ELISA to quantitate PSRC1 in samples. An antibody specific for PSRC1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PSRC1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PSRC1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PSRC1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: PSRC1 encodes a proline-rich protein. Studies of the related mouse gene suggest that this gene is regulated by p53 and may participate in p53-mediated growth suppression. Alternatively spliced transcript variants encoding different isoforms have been described. The deduced 333-amino acid protein shares 68.2% identity with mouse Dda3, with highest conservation in the N-terminal half. Both mouse and human DDA3 are rich in serine and proline and contain a coiled-coil domain, and 3 of 6 PxxP motifs in mouse Dda3 are conserved in human DDA3. PCR analysis revealed variable DDA3 expression in all human tissues examined except skeletal muscle. Highest expression was detected in fetal thymus and adult brain.

NCBI Description

This gene encodes a proline-rich protein that is a target for regulation by the tumor suppressor protein p53. The encoded protein plays an important role in mitosis by recruiting and regulating microtubule depolymerases that destabalize microtubules. Alternatively spliced transcript variants encoding different isoforms have been described. [provided by RefSeq, Apr 2014]

Required for normal progression through mitosis. Required for normal congress of chromosomes at the metaphase plate, and for normal rate of chromosomal segregation during anaphase. Plays a role in the regulation of mitotic spindle dynamics. Increases the rate of turnover of microtubules on metaphase spindles, and contributes to the generation of normal tension across sister kinetochores. Recruits KIF2A and ANKRD53 to the mitotic spindle and spindle poles. May participate in p53/TP53-regulated growth suppression.
