Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate HERC1 in samples. An antibody specific for HERC1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyHERC1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HERC1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HERC1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

HERC1: Involved in membrane trafficking via some guanine nucleotide exchange factor (GEF) activity and its ability to bind clathrin. Acts as a GEF for Arf and Rab, by exchanging bound GDP for free GTP. Binds phosphatidylinositol 4,5-bisphosphate, which is required for GEF activity. May also act as a E3 ubiquitin- protein ligase which accepts ubiquitin from an E2 ubiquitin- conjugating enzyme in the form of a thioester and then directly transfers the ubiquitin to targeted substrates.
