Principle of the Assay: The Picokine™ Human PRELP Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human PRELP with a 96-well strip plate that is pre-coated with antibody specific for PRELP. The detection antibody is a biotinylated antibody specific for PRELP. The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated monoclonal antibody from goat. The kit contains recombinant Human PRELP with immunogen: Expression system for standard: NSO, Immunogen sequence: Q21-I382. The kit is analytically validated with ready to use reagents. To measure Human PRELP, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Human PRELP in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human PRELP in the sample.!!Background/Introduction: Prolargin is a protein that in humans is encoded by the PRELP gene. It is mapped to 1q32.1. The protein encoded by this gene is a leucine-rich repeat protein present in connective tissue extracellular matrix. This protein functions as a molecule anchoring basement membranes to the underlying connective tissue. This protein has been shown to bind type I collagen to basement membranes and type II collagen to cartilage. It also binds the basement membrane heparan sulfate proteoglycan perlecan. This protein is suggested to be involved in the pathogenesis of Hutchinson- Gilford progeria (HGP), which is reported to lack the binding of collagen in basement membranes and cartilage. Alternatively spliced transcript variants encoding the same protein have been observed.
