The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Placental Thrombin Inhibitor (PTI). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Placental Thrombin Inhibitor (PTI). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Placental Thrombin Inhibitor (PTI), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Placental Thrombin Inhibitor (PTI) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

SERPINB6: May be involved in the regulation of serine proteinases present in the brain or extravasated from the blood. Inhibitor of cathepsin G, kallikrein-8 and thrombin. May play an important role in the inner ear in the protection against leakage of lysosomal content during stress and loss of this protection results in cell death and sensorineural hearing loss. Defects in SERPINB6 are the cause of deafness autosomal recessive type 91 (DFNB91). It is a form of non- syndromic deafness characterized by progressive and age-dependent sensorineural hearing loss. Vestibular function is normal. Belongs to the serpin family. Ov-serpin subfamily.
