Principle of the Assay: PlGF ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-PlGF antibody and an PlGF-HRP conjugate. The assay sample and buffer are incubated together with PlGF-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PlGF concentration since PlGF from samples and PlGF-HRP conjugate compete for the anti-PlGF antibody binding site. Since the number of sites is limited, as more sites are occupied by PlGF from the sample, fewer sites are left to bind PlGF-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PlGF concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is found as a pentamer and is a major substrate for the cAMP-dependent protein kinase in cardiac muscle. The encoded protein is an inhibitor of cardiac muscle sarcoplasmic reticulum Ca(2+)-ATPase in the unphosphorylated state, but inhibition is relieved upon phosphorylation of the protein. The subsequent activation of the Ca(2+) pump leads to enhanced muscle relaxation rates, thereby contributing to the inotropic response elicited in heart by beta-agonists. The encoded protein is a key regulator of cardiac diastolic function. Mutations in this gene are a cause of inherited human dilated cardiomyopathy with refractory congestive heart failure.
