Intended Uses: This PlGF ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat PlGF. This ELISA kit for research use only!

Principle of the Assay: PlGF ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-PlGF antibody and an PlGF-HRP conjugate. The assay sample and buffer are incubated together with PlGF-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PlGF concentration since PlGF from samples and PlGF-HRP conjugate compete for the anti-PlGF antibody binding site. Since the number of sites is limited, as more sites are occupied by PlGF from the sample, fewer sites are left to bind PlGF-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PlGF concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is found as a pentamer and is a major substrate for the cAMP-dependent protein kinase in cardiac muscle. The encoded protein is an inhibitor of cardiac muscle sarcoplasmic reticulum Ca(2+)-ATPase in the unphosphorylated state, but inhibition is relieved upon phosphorylation of the protein. The subsequent activation of the Ca(2+) pump leads to enhanced muscle relaxation rates, thereby contributing to the inotropic response elicited in heart by beta-agonists. The encoded protein is a key regulator of cardiac diastolic function. Mutations in this gene are a cause of inherited human dilated cardiomyopathy with refractory congestive heart failure. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Phospholamban has been postulated to regulate the activity of the calcium pump of cardiac sarcoplasmic reticulum.

Subunit structure: Homopentamer. Interacts with HAX1. Ref.8

Subcellular location: Mitochondrion membrane; Single-pass membrane protein. Sarcoplasmic reticulum

By similarity.

Tissue specificity: Heart.

Post-translational modification: Phosphorylated at Thr-17 by CaMK2, and in response to beta-adrenergic stimulation. Phosphorylation by DMPK may stimulate sarcoplasmic reticulum calcium uptake in cardiomyocytes. Ref.6 Ref.7

Involvement in disease: Cardiomyopathy, dilated 1P (CMD1P) [MIM:609909]: A disorder characterized by ventricular dilation and impaired systolic function, resulting in congestive heart failure and arrhythmia. Patients are at risk of premature death.Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.12 Ref.13Cardiomyopathy, familial hypertrophic 18 (CMH18) [MIM:613874]: A hereditary heart disorder characterized by ventricular hypertrophy, which is usually asymmetric and often involves the interventricular septum. The symptoms include dyspnea, syncope, collapse, palpitations, and chest pain. They can be readily provoked by exercise. The disorder has inter- and intrafamilial variability ranging from benign to malignant forms with high risk of cardiac failure and sudden cardiac death.Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.5

Sequence similarities: Belongs to the phospholamban family.

Product Notes

The Human PLN pln (Catalog #AAA726922) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA726922 ELISA Kit recognizes Human PLN. It is sometimes possible for the material contained within the vial of "phospholamban, PLN, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
