Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for PTH elisa kit
Description: This Human Parathyroid hormone (PTH) ELISA Kit employs a two-site sandwich ELISA to quantitate PTH in samples. An antibody specific for PTH has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PTH present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PTH is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PTH bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Parathyroid hormone is secreted by the parathyroid glands as a polypeptide containing 84 amino acids. It acts to increase the concentration of calcium (Ca2+) in the blood, whereas calcitonin (a hormone produced by the parafollicular cells (C cells) of the thyroid gland) acts to decrease calcium concentration. PTH acts to increase the concentration of calcium in the blood by acting upon parathyroid hormone receptor in three parts of the body: PTH half-life is approximately 4 minutes. It has a molecular mass of 9.4 kDa.PTH was one of the first hormones to be shown to use the G-protein, adenylyl cyclase second messenger system.Normal total plasma calcium level ranges from 8.5 to 10.2 mg/dL (2.12 mmol/L to 2.55 mmol/L).

NCBI Description

This gene encodes a member of the parathyroid family of proteins. The encoded preproprotein is proteolytically processed to generate a protein that binds to the parathyroid hormone/parathyroid hormone-related peptide receptor and regulates blood calcium and phosphate levels. Excess production of the encoded protein, known as hyperparathyroidism, can result in hypercalcemia and kidney stones. On the other hand, defective processing of the encoded protein may lead to hypoparathyroidism, which can result in hypocalcemia and numbness. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Oct 2015]

Uniprot Description

PTH: PTH elevates calcium level by dissolving the salts in bone and preventing their renal excretion. Stimulates [1-14C]-2- deoxy-D-glucose (2DG) transport and glycogen synthesis in osteoblastic cells. Defects in PTH are a cause of familial isolated hypoparathyroidism (FIH); also called autosomal dominant hypoparathyroidism or autosomal dominant hypocalcemia. FIH is characterized by hypocalcemia and hyperphosphatemia due to inadequate secretion of parathyroid hormone. Symptoms are seizures, tetany and cramps. FIH exist both as autosomal dominant and recessive forms of hypoparathyroidism. Belongs to the parathyroid hormone family.

Product Notes

The Human PTH pth (Catalog #AAA9716277) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9716277 ELISA Kit recognizes Human PTH. It is sometimes possible for the material contained within the vial of "Parathyroid Hormone (PTH), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
