Principle of the Assay: OR4D11 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-OR4D11 antibody and an OR4D11-HRP conjugate. The assay sample and buffer are incubated together with OR4D11-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the OR4D11 concentration since OR4D11 from samples and OR4D11-HRP conjugate compete for the anti-OR4D11 antibody binding site. Since the number of sites is limited, as more sites are occupied by OR4D11 from the sample, fewer sites are left to bind OR4D11-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The OR4D11 concentration in each sample is interpolated from this standard curve.

The olfactory receptor proteins are members of a large family of G-protein-coupled receptors (GPCR) arising from single coding-exon genes. Olfactory receptors share a 7-transmembrane domain structure with many neurotransmitter and hormone receptors and are responsible for the recognition and G protein-mediated transduction of odorant signals. The olfactory receptor gene family is the largest in the genome. The nomenclature assigned to the olfactory receptor genes and proteins for this organism is independent of other organisms.
