The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to NLR Family, Pyrin Domain Containing Protein 7 (NLRP7). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to NLR Family, Pyrin Domain Containing Protein 7 (NLRP7). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain NLR Family, Pyrin Domain Containing Protein 7 (NLRP7), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of NLR Family, Pyrin Domain Containing Protein 7 (NLRP7) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

This gene encodes a member of the NACHT, leucine rich repeat, and PYD containing (NLRP) protein family. It has an N-terminal pyrin domain, followed by a NACHT domain, a NACHT-associated domain (NAD), and a C-terminal leucine-rich repeat (LRR) region. NLRP proteins are implicated in the activation of proinflammatory caspases through multiprotein complexes called inflammasomes. This gene may act as a feedback regulator of caspase-1-dependent interleukin 1-beta secretion. Alternative splicing results in multiple transcript variants encoding different isoforms. NLRP7: Inhibits CASP1/caspase-1-dependent IL1B secretion. Defects in NLRP7 are the cause of hydatidiform mole recurrent type 1 (HYDM1). A disorder characterized by excessive trophoblast development and the presence of a growing mass of tissue inside the uterus at the beginning of a pregnancy. It leads to abnormal pregnancies with no embryo, and cystic degeneration of the chorionic villi. Belongs to the NLRP family. 3 isoforms of the human protein are produced by alternative splicing.
