Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for NAE1 elisa kit
Description: This Human NEDD8-activating enzyme E1 regulatory subunit (NAE1) ELISA Kit employs a two-site sandwich ELISA to quantitate NAE1 in samples. An antibody specific for NAE1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any NAE1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NAE1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NAE1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: NEDD8-activating enzyme E1 regulatory subunit encoded by NAE1 binds to the beta-amyloid precursor protein. Beta-amyloid precursor protein is a cell surface protein with signal-transducing properties, and it is thought to play a role in the pathogenesis of Alzheimer's disease. In addition, the encoded protein can form a heterodimer with UBE1C and bind and activate NEDD8, a ubiquitin-like protein. This protein is required for cell cycle progression through the S/M checkpoint. Three transcript variants encoding different isoforms have been found for NAE1.

Uniprot Description

APPBP1: Regulatory subunit of the dimeric UBA3-NAE1 E1 enzyme. E1 activates NEDD8 by first adenylating its C-terminal glycine residue with ATP, thereafter linking this residue to the side chain of the catalytic cysteine, yielding a NEDD8-UBA3 thioester and free AMP. E1 finally transfers NEDD8 to the catalytic cysteine of UBE2M. Necessary for cell cycle progression through the S-M checkpoint. Overexpression of NAE1 causes apoptosis through deregulation of NEDD8 conjugation. Belongs to the ubiquitin-activating E1 family. ULA1 subfamily. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Ubiquitin conjugating system

Chromosomal Location of Human Ortholog: 16q22

Product Notes

The Human NAE1 nae1 (Catalog #AAA9715055) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9715055 ELISA Kit recognizes Human NAE1. It is sometimes possible for the material contained within the vial of "NEDD8-Activating Enzyme E1 Regulatory Subunit (NAE1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
