Principle of the Assay: Human Myelin-Associated Glycoprotein (MAG) ELISA Kit employs a two-site sandwich ELISA to quantitate MAG in samples. An antibody specific for MAG has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MAG present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human MAG detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of MAG bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background/Introduction: Myelin-associated glycoprotein (MAG) is a type 1 transmembrane protein glycoprotein localized in periaxonal Schwann cell and oligodendrocyte membranes, where it plays a role in glial-axonal interactions. MAG is a member of the SIGLEC family of proteins and is a functional ligand of the NOGO-66 receptor, NgR. MAG is believed to be involved in myelination during nerve regeneration in the PNS and is vital for the long-term survival of the myelinated axons following myelinogenesis. In the CNS MAG is one of three main myelin-associated inhibitors of axonal regeneration after injury, making it an important protein for future research on neurogenesis in the CNS.

NCBI Description

The protein encoded by this gene is a type I membrane protein and member of the immunoglobulin superfamily. It is thought to be involved in the process of myelination. It is a lectin that binds to sialylated glycoconjugates and mediates certain myelin-neuron cell-cell interactions. Three alternatively spliced transcripts encoding different isoforms have been described for this gene. [provided by RefSeq, Nov 2010]

Uniprot Description

MAG: Adhesion molecule in postnatal neural development that mediates sialic-acid dependent cell-cell interactions between neuronal and myelinating cells. Preferentially binds to alpha-2,3- linked sialic acid. Binds to RTN4R. Belongs to the immunoglobulin superfamily. SIGLEC (sialic acid binding Ig-like lectin) family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Cell adhesion; Membrane protein, integral

Chromosomal Location of Human Ortholog: 19q13.1

Cellular Component: paranode region of axon; integral to membrane; plasma membrane

Molecular Function: carbohydrate binding

Biological Process: substantia nigra development; nerve growth factor receptor signaling pathway; negative regulation of axonogenesis; regulation of axonogenesis; blood coagulation; cell adhesion; leukocyte migration

Product Notes

The Human MAG mag (Catalog #AAA9715415) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9715415 ELISA Kit recognizes Human MAG. It is sometimes possible for the material contained within the vial of "Myelin-Associated Glycoprotein (MAG), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
