Principle of the Assay: Human Mutated Citrullinated Vimentin (MCV) ELISA Kit employs a two-site sandwich ELISA to quantitate MCV in samples. An antibody specific for MCV has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MCV present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human MCV detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of MCV bound in the initial step. The color development is stopped and the intensity of the color is measured.
Background/Introduction: Autoantibodies against mutated and citrullinated vimentin (MCV) represent a novel test marker for rheumatoid arthritis (RA). Recently, an increased sensitivity for anti-MCV compared to autoantibodies against cyclic citrullinated peptides (anti-CCP2) was shown in cohorts of patients with early RA and established disease. The aim of this study was to develop and evaluate a point of care test (POCT) for detection of anti-MCV antibodies immediately at the first visit or at the bed side.
