Human Mitotic Spindle Assembly Checkpoint Protein MAD2B (MAD2L2) ELISA Kit | MAD2L2 elisa kit
Human Mitotic Spindle Assembly Checkpoint Protein MAD2B (MAD2L2) ELISA Kit
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Background: MAD2L2 is a component of the mitotic spindle assembly checkpoint that prevents the onset of anaphase until all chromosomes are properly aligned at the metaphase plate. MAD2L2 is a homolog of MAD2L1. The predicted 211-amino acid MAD2B protein shares 24% and 26% sequence identity with yeast Mad2 and human MAD2L1, respectively, in the conserved regions. RT-PCR analysis revealed that both human MAD2 homologs were expressed at similar high levels in a panel of cell lines. Further binding analyses determined that the interaction of MAD2L2 with ADAM9 is mediated through a proline-rich SH3-ligand domain of ADAM9. Northern blot analysis detected 1.35-kb MAD2L3 transcripts in all tissues tested, with highest expression in testis.
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is a component of the mitotic spindle assembly checkpoint that prevents the onset of anaphase until all chromosomes are properly aligned at the metaphase plate. The encoded protein, which is similar to MAD2L1, is capable of interacting with ADAM9, ADAM15, REV1, and REV3 proteins. [provided by RefSeq, Jul 2008]
Uniprot Description
MAD2L2: Adapter protein able to interact with different proteins and involved in different biological processes. Mediates the interaction between the error-prone DNA polymerase zeta catalytic subunit REV3L and the inserter polymerase REV1, thereby mediating the second polymerase switching in translesion DNA synthesis. Translesion DNA synthesis releases the replication blockade of replicative polymerases, stalled in presence of DNA lesions. May also regulate another aspect of cellular response to DNA damage through regulation of the JNK-mediated phosphorylation and activation of the transcriptional activator ELK1. Inhibits the FZR1- and probably CDC20-mediated activation of the anaphase promoting complex APC thereby regulating progression through the cell cycle. Regulates TCF7L2-mediated gene transcription and may play a role in epithelial-mesenchymal transdifferentiation.

Product Notes
The Human MAD2L2 mad2l2 (Catalog #AAA9715421) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9715421 ELISA Kit recognizes Human MAD2L2. It is sometimes possible for the material contained within the vial of "Mitotic Spindle Assembly Checkpoint Protein MAD2B (MAD2L2), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
