Uniprot Description

Function: Dual specificity protein kinase which acts as an essential component of the MAP kinase signal transduction pathway. Catalyzes the concomitant phosphorylation of a threonine and a tyrosine residue in the MAP kinases p38 and plays an important role in the regulation of cellular responses to cytokines and all kinds of stresses. The p38 MAP kinase signal transduction pathway leads to direct activation of transcription factors. Phosphorylation by MAP2K6 asymmetrically activates p38 on one side of the blastodisc, an event which is necessary for blastomere cleavage. Ref.1

Catalytic activity: ATP + a protein = ADP + a phosphoprotein.

Enzyme regulation: Activated by dual phosphorylation on Ser-234 and Thr-238 in response to a variety of cellular stresses, including UV radiation, osmotic shock, hypoxia, inflammatory cytokines, interferon gamma (IFNG), and less often by growth factors. MAP2K6/MKK6 is activated by the majority of M3Ks

By similarity.

Subunit structure: Dimer

By similarity. Interacts (via its D domain) with its MAP kinase substrates

By similarity. Interacts (via its DVD domain) with MAP3Ks activators

By similarity.

Subcellular location: Nucleus

By similarity. Cytoplasm

By similarity. Cytoplasm › cytoskeleton

By similarity. Note: Binds to microtubules

By similarity.

Developmental stage: Expressed both maternally and zygotically. Zygotic expression continues throughout development. Ref.1

Domain: The DVD domain (residues 338-361) contains a conserved docking site and is found in the mammalian MAP kinase kinases (MAP2Ks). The DVD sites bind to their specific upstream MAP kinase kinase kinases (MAP3Ks) and are essential for activation

By similarity. The D domain (residues 30-46) contains a conserved docking site and is required for the binding to MAPK substrates

By similarity.

Post-translational modification: Weakly autophosphorylated

By similarity. Phosphorylated at Ser-234 and Thr-238 by the majority of M3Ks.

Sequence similarities: Belongs to the protein kinase superfamily. STE Ser/Thr protein kinase family. MAP kinase kinase subfamily.Contains 1 protein kinase domain.

Caution: Was originally (Ref.1) thought to be a Map2k3 protein but sequence analysis shows closer similarity to the Map2k6 proteins.

Sequence caution: The sequence AAH71285.1 differs from that shown. Reason: Erroneous initiation. Translation N-terminally extended.

Product Notes

The Human MAP2K6 map2k6 (Catalog #AAA9304470) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9304470 ELISA Kit recognizes Human MAP2K6. It is sometimes possible for the material contained within the vial of "Mitogen Activated Protein Kinase Kinase 6 (MAP2K6), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
