6. Aspirate and wash 5 times
7. Add 90ul Substrate Solution. Incubate 10-20 minutes at 37 degree C
8. Add 50ul Stop Solution. Read at 450nm immediately.
Test Principle
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Glutathione Peroxidase 1 (GPX1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Glutathione Peroxidase 1 (GPX1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Glutathione Peroxidase 1 (GPX1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Glutathione Peroxidase 1 (GPX1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
Preparation and Storage
Store at 4 degree C for 1 month/ -20 degree C for long-term (within shelf life).
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

NCBI Description

The liver X receptors, LXRA (NR1H3; MIM 602423) and LXRB, form a subfamily of the nuclear receptor superfamily and are key regulators of macrophage function, controlling transcriptional programs involved in lipid homeostasis and inflammation. The inducible LXRA is highly expressed in liver, adrenal gland, intestine, adipose tissue, macrophages, lung, and kidney, whereas LXRB is ubiquitously expressed. Ligand-activated LXRs form obligate heterodimers with retinoid X receptors (RXRs; see MIM 180245) and regulate expression of target genes containing LXR response elements (summary by Korf et al., 2009 [PubMed 19436111]).[supplied by OMIM, Jan 2010]

Uniprot Description

LXR-beta: Orphan receptor. Binds preferentially to double-stranded oligonucleotide direct repeats having the consensus half-site sequence 5'-AGGTCA-3' and 4-nt spacing (DR-4). Regulates cholesterol uptake through MYLIP-dependent ubiquitination of LDLR, VLDLR and LRP8. Forms a heterodimer with RXR. Ubiquitous. Belongs to the nuclear hormone receptor family. NR1 subfamily.

Product Notes

The Human LXRb nr1h2 (Catalog #AAA2709685) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA2709685 ELISA Kit recognizes Human LXRb. AAA Biotech's Liver X Receptor Beta (LXRb) can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). Enzyme-linked immunosorbent assay for Antigen Detection. Researchers should empirically determine the suitability of the LXRb nr1h2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Liver X Receptor Beta (LXRb), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
