Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Lipocalin-1 (LCN1) ELISA Kit employs a two-site sandwich ELISA to quantitate LCN1 in samples. An antibody specific for LCN1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any LCN1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LCN1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LCN1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Lipocalin-1 belongs to the lipocalin family. Lipocalins are a group of extracellular proteins that are able to bind lipophiles by enclosure within their structures to minimize solvent contact. This protein may bind hydrophobic ligands and inhibit cysteine proteinases. It may also play a role in taste reception. Based on the known 3-dimensional structure of 5 members of the lipocalin family, i.e., retinol binding protein, beta-lactoglobulin, bilin binding protein, mouse major urinary protein, and rat urinary alpha-2-globulin, the general architecture appears to be highly appropriate for binding a variety of hydrophobic ligands. On the basis of highly conserved amino acid sequences and of a size around 18 to 20 kD, about 20 proteins have been designated as lipocalins.

NCBI Description

This gene encodes a member of the lipocalin family of small secretory proteins. Lipocalins are extracellular transport proteins that bind to a variety of hydrophobic ligands. The encoded protein is the primary lipid binding protein in tears and is overproduced in response to multiple stimuli including infection and stress. The encoded protein may be a marker for chromosome aneuploidy as well as an autoantigen in Sjogren's syndrome. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene, and two pseudogenes of this gene are also located on the long arm of chromosome 9.
