Principle of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human HIST1H1C antibody. HIST1H1C present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human HIST1H1C Antibody is added and binds to HIST1H1C in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated HIST1H1C antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human HIST1H1C. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Histones are basic nuclear proteins responsible for nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. This gene is intronless and encodes a replication-dependent histone that is a member of the histone H1 family. Transcripts from this gene lack polyA tails but instead contain a palindromic termination element. This gene is found in the large histone gene cluster on chromosome 6. [provided by RefSeq, Aug 2015]

Histones H1 are necessary for the condensation of nucleosome chains into higher-order structured fibers.
