Intended Uses: This sandwich kit is for the accurate quantitative detection of human Growth Arrest and DNA Damage Inducible Protein Alpha (also known as GADD45a) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human GADD45a antibody. GADD45a present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human GADD45a Antibody is added and binds to GADD45a in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated GADD45a antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human GADD45a. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

NCBI Description

This gene is a member of a group of genes whose transcript levels are increased following stressful growth arrest conditions and treatment with DNA-damaging agents. The protein encoded by this gene responds to environmental stresses by mediating activation of the p38/JNK pathway via MTK1/MEKK4 kinase. The DNA damage-induced transcription of this gene is mediated by both p53-dependent and -independent mechanisms. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene.[provided by RefSeq, Dec 2010]

In T-cells, functions as a regulator of p38 MAPKs by inhibiting p88 phosphorylation and activity (). Might affect PCNA interaction with some CDK (cell division protein kinase) complexes; stimulates DNA excision repair in vitro and inhibits entry of cells into S phase.
