Product Information: Granzyme B (human) ELISA Kit: Colorimetric Assay for Quantitative measurement of human Granzyme B in serum, plasma & cell culture supernatants.Sensitivity: < 10 pg/ml. 100 assays.

Background: Granzyme B is a serine protease expressed by cytotoxic T lymphocytes (CTL) and natural killer (NK) cells and functions in cell-mediated immunity. MyBioSource's human Granzyme B ELISA Kit is based on the standard principle of a sandwich enzyme-linked immunosorbent assay. A mouse monoclonal antibody specific for human Granzyme B is coated on a 96-well plate. Standards and test samples are added to the wells and Granzyme B present in a sample is bound by the immobilized antibody. A biotinylated polyclonal antibody from goat specific for Granzyme B is added subsequently. After washing away the unbound biotinylated antibody with PBS or TBS buffer, avidin-Biotin-Peroxidase Complex is added to the wells. The wells are again washed with PBS or TBS buffer to remove the unbound conjugates. HRP substrate TMB is used to visualize the HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue product that changes into yellow after adding acidic stop solution. The density of yellow color is proportional to the human Granzyme B captured onto the plate. This ELISA kit shows no cross-reactivity with other relevant proteins. Detection Range: 15.6 pg/ml - 1000 pg/ml. Sensitivity: < 10 pg/ml.

NCBI Description

Cytolytic T lymphocytes (CTL) and natural killer (NK) cells share the remarkable ability to recognize, bind, and lyse specific target cells. They are thought to protect their host by lysing cells bearing on their surface 'nonself' antigens, usually peptides or proteins resulting from infection by intracellular pathogens. The protein encoded by this gene is crucial for the rapid induction of target cell apoptosis by CTL in cell-mediated immune response. [provided by RefSeq, Jul 2008]

Uniprot Description

GZMB: This enzyme is necessary for target cell lysis in cell- mediated immune responses. It cleaves after Asp. Seems to be linked to an activation cascade of caspases (aspartate-specific cysteine proteases) responsible for apoptosis execution. Cleaves caspase-3, -7, -9 and 10 to give rise to active enzymes mediating apoptosis. By staphylococcal enterotoxin A (SEA) in peripheral blood leukocytes. Inactivated by the serine protease inhibitor diisopropylfluorophosphate. Belongs to the peptidase S1 family. Granzyme subfamily.

Product Notes

The Human GZMB gzmb (Catalog #AAA843467) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA843467 ELISA Kit recognizes Human GZMB. Application- Quantitative protein detection, establishing normal range. Researchers should empirically determine the suitability of the GZMB gzmb for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Granzyme B, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
