Intended Uses: This sandwich kit is for the accurate quantitative detection of Human Glutathione Peroxidase 3 (also known as GPX3) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Human GPX3 antibody. GPX3 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human GPX3 Antibody is added and binds to GPX3 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated GPX3 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human GPX3. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

NCBI Description

The protein encoded by this gene belongs to the glutathione peroxidase family, members of which catalyze the reduction of organic hydroperoxides and hydrogen peroxide (H2O2) by glutathione, and thereby protect cells against oxidative damage. Several isozymes of this gene family exist in vertebrates, which vary in cellular location and substrate specificity. This isozyme is secreted and is predominantly expressed in rat kidney, which appears to be the major source of the enzyme in plasma. Dysregulation of this isozyme has been associated with obesity-related metabolic complications. This isozyme is also a selenoprotein, containing the rare amino acid selenocysteine (Sec) at its active site. Sec is encoded by the UGA codon, which normally signals translation termination. The 3' UTRs of selenoprotein mRNAs contain a conserved stem-loop structure, designated the Sec insertion sequence (SECIS) element, that is necessary for the recognition of UGA as a Sec codon, rather than as a stop signal.
