Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Galanin (GAL) ELISA Kit employs a two-site sandwich ELISA to quantitate GAL in samples. An antibody specific for GAL has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any GAL present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for GAL is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GAL bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Galanin is formed by the cleavage of a prepropeptide encoded by a gene known as GAL. It is involved in a number of physiological processes such as regulation of food intake, metabolism and reproduction, regulation of neurotransmitter and hormone release, nociception, intestinal contraction and secretion, and more recently in nervous system development and response to injury. This wide diversity of action is mediated by several galanin receptor subtypes.Galanin is predominantly an inhibitory, hyperpolarizing neuropeptide and as such inhibits neurotransmitter release. Galanin is often co-localized with classical neurotransmitters such as acetylcholine, serotonin and norepinephrine and also with other neuromodulators such as Neuropeptide Y, Substance P and Vasoactive intestinal peptide.

NCBI Description

This gene encodes a neuroendocrine peptide that is widely expressed in the central and peripheral nervous systems and also the gastrointestinal tract, pancreas, adrenal gland and urogenital tract. The encoded protein is a precursor that is proteolytically processed to generate two mature peptides: galanin and galanin message-associated peptide (GMAP). Galanin has diverse physiological functions including nociception, feeding and energy homeostasis, osmotic regulation and water balance. GMAP has been demonstrated to possess antifungal activity and hypothesized to be part of the innate immune system. [provided by RefSeq, Jul 2015]
