Human F box only protein 41 (FBXO41) ELISA Kit
Principle of the Assay: FBXO41 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-FBXO41 antibody and an FBXO41-HRP conjugate. The assay sample and buffer are incubated together with FBXO41-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FBXO41 concentration since FBXO41 from samples and FBXO41-HRP conjugate compete for the anti-FBXO41 antibody binding site. Since the number of sites is limited, as more sites are occupied by FBXO41 from the sample, fewer sites are left to bind FBXO41-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FBXO41 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the F-box protein family, which is characterized by an approximately 40 amino acid motif, the F-box. F-box proteins constitute one of the four subunits of the SCF ubiquitin protein ligase complex that plays a role in phosphorylation-dependent ubiquitination. F-box proteins are divided into three classes depending on the interaction substrate domain each contains in addition to the F-box motif: FBXW proteins contain WD-40 domains, FBXL proteins contain leucine-rich repeats, and FBXO proteins contain either different protein-protein interaction modules or no recognizable motifs.
