Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for XPO5 elisa kit
Description: This Human Exportin-5 (XPO5) ELISA Kit employs a two-site sandwich ELISA to quantitate XPO5 in samples. An antibody specific for XPO5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any XPO5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for XPO5 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of XPO5 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Exportin-5 belongs to a large family of karyopherins that mediate the transport of proteins and other cargo between the nuclear and cytoplasmic compartments. The deduced 1,205-amino acid protein has a calculated molecular mass of about 136 kD. XPO5 and XPO1 share 25% amino acid identity. Northern blot analysis detected transcripts of about 3.6 and 5.0 kb in all tissues examined.XPO5 bound the nucleoporins NUP214 and NUP153 in a RAN-GTP-independent manner and bound a cargo protein, ILF3, in a RAN-GTP-dependent manner. XPO5 bound ILF3 at its double-stranded RNA-binding domain (dsRBD) and also bound the dsRBDs of other proteins. XPO5 directly interacted with VA1 RNA in a RAN-GTP-dependent manner.

NCBI Description

This gene encodes a member of the karyopherin family that is required for the transport of small RNAs and double-stranded RNA-binding proteins from the nucleus to the cytoplasm. The encoded protein translocates cargo through the nuclear pore complex in a RanGTP-dependent process. [provided by RefSeq, Aug 2011]

Uniprot Description

Mediates the nuclear export of proteins bearing a double-stranded RNA binding domain (dsRBD) and double-stranded RNAs (cargos). XPO5 in the nucleus binds cooperatively to the RNA and to the GTPase Ran in its active GTP-bound form. Proteins containing dsRBDs can associate with this trimeric complex through the RNA. Docking of this complex to the nuclear pore complex (NPC) is mediated through binding to nucleoporins. Upon transit of a nuclear export complex into the cytoplasm, hydrolysis of Ran-GTP to Ran-GDP (induced by RANBP1 and RANGAP1, respectively) cause disassembly of the complex and release of the cargo from the export receptor. XPO5 then returns to the nuclear compartment by diffusion through the nuclear pore complex, to mediate another round of transport. The directionality of nuclear export is thought to be conferred by an asymmetric distribution of the GTP- and GDP-bound forms of Ran between the cytoplasm and nucleus. Overexpression may in some circumstances enhance RNA-mediated gene silencing (RNAi). Mediates nuclear export of isoform 5 of ADAR/ADAR1 in a RanGTP-dependent manner.

Product Notes

The Human XPO5 xpo5 (Catalog #AAA9713680) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9713680 ELISA Kit recognizes Human XPO5. It is sometimes possible for the material contained within the vial of "Exportin-5 (XPO5), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
