Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Dual specificity protein kinase TTK (TTK) ELISA Kit employs a two-site sandwich ELISA to quantitate TTK in samples. An antibody specific for TTK has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TTK present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TTK is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TTK bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: This au protein kinase belongs to the family of transferases, specifically those transferring a phosphate group to the sidechain oxygen atom of serine or threonine residues in proteins.This enzyme participates in 14 metabolic pathways: erbb signaling pathway, cell cycle, wnt signaling pathway, hedgehog signaling pathway, axon guidance, focal adhesion, b cell receptor signaling pathway, insulin signaling pathway, melanogenesis, alzheimer's disease, colorectal cancer, endometrial cancer, prostate cancer, and basal cell carcinoma.Expressed in the brain, particularly in cortical and hippocampal neurons. Weakly expressed in spinal cord and testis. No expression in adipose tissue, bladder, cervix, colon, esophagus, heart, kidney, liver, lung, ovary, placenta, prostate, skeletal muscle, small intestine, spleen, testis, thymus, thyroid or trachea.

NCBI Description

This gene encodes a dual specificity protein kinase with the ability to phosphorylate tyrosine, serine and threonine. Associated with cell proliferation, this protein is essential for chromosome alignment at the centromere during mitosis and is required for centrosome duplication. It has been found to be a critical mitotic checkpoint protein for accurate segregation of chromosomes during mitosis. Tumorigenesis may occur when this protein fails to degrade and produces excess centrosomes resulting in aberrant mitotic spindles. Alternative splicing results in multiple transcript variants. Uniprot Description

TTK: a dual-specificity protein kinase orthologous to the yeast monopolar spindle (Mps1) kinase that is required for spindle pole duplication and the spindle checkpoint but not for centrosome duplication. Localized to centrosomes, kinetochores and nuclear pores. Its enzymatic activity is required for the recruitment of Mad1, Mad2, Bub1 and Bub3 to the kinetochore. Mps1 mRNA levels change little throughout the cell cycle, but its protein expression varies during the cell cycle. Overexpression arrests cells in metaphase by activating the checkpoint.
