This assay has high sensitivity and excellent specificity for detection of COX7C. No significant cross-reactivity or interference between COX7C and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between COX7C and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This COX7C ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human COX7C. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: COX7C ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-COX7C antibody and an COX7C-HRP conjugate. The assay sample and buffer are incubated together with COX7C-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the COX7C concentration since COX7C from samples and COX7C-HRP conjugate compete for the anti-COX7C antibody binding site. Since the number of sites is limited, as more sites are occupied by COX7C from the sample, fewer sites are left to bind COX7C-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The COX7C concentration in each sample is interpolated from this standard curve.

NCBI Description

Cytochrome c oxidase (COX), the terminal component of the mitochondrial respiratory chain, catalyzes the electron transfer from reduced cytochrome c to oxygen. This component is a heteromeric complex consisting of 3 catalytic subunits encoded by mitochondrial genes and multiple structural subunits encoded by nuclear genes. The mitochondrially-encoded subunits function in electron transfer, and the nuclear-encoded subunits may function in the regulation and assembly of the complex. This nuclear gene encodes subunit VIIc, which shares 87% and 85% amino acid sequence identity with mouse and bovine COX VIIc, respectively, and is found in all tissues. A pseudogene COX7CP1 has been found on chromosome 13. [provided by RefSeq, Jul 2008]
