Principle of the Assay: COX7C ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-COX7C antibody and an COX7C-HRP conjugate. The assay sample and buffer are incubated together with COX7C-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the COX7C concentration since COX7C from samples and COX7C-HRP conjugate compete for the anti-COX7C antibody binding site. Since the number of sites is limited, as more sites are occupied by COX7C from the sample, fewer sites are left to bind COX7C-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The COX7C concentration in each sample is interpolated from this standard curve.

Cytochrome c oxidase (COX), the terminal component of the mitochondrial respiratory chain, catalyzes the electron transfer from reduced cytochrome c to oxygen. This component is a heteromeric complex consisting of 3 catalytic subunits encoded by mitochondrial genes and multiple structural subunits encoded by nuclear genes. The mitochondrially-encoded subunits function in electron transfer, and the nuclear-encoded subunits may function in the regulation and assembly of the complex. This nuclear gene encodes subunit VIIc, which shares 87% and 85% amino acid sequence identity with mouse and bovine COX VIIc, respectively, and is found in all tissues.
