Human Cysteine protease ATG4B (ATG4B) ELISA Kit
Principle of the Assay: ATG4B ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ATG4B antibody and an ATG4B-HRP conjugate. The assay sample and buffer are incubated together with ATG4B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ATG4B concentration since ATG4B from samples and ATG4B-HRP conjugate compete for the anti-ATG4B antibody binding site. Since the number of sites is limited, as more sites are occupied by ATG4B from the sample, fewer sites are left to bind ATG4B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ATG4B concentration in each sample is interpolated from this standard curve.

Autophagy is the process by which endogenous proteins and damaged organelles are destroyed intracellularly. Autophagy is postulated to be essential for cell homeostasis and cell remodeling during differentiation, metamorphosis, non-apoptotic cell death, and aging. Reduced levels of autophagy have been described in some malignant tumors, and a role for autophagy in controlling the unregulated cell growth linked to cancer has been proposed. This gene encodes a member of the autophagin protein family. The encoded protein is also designated as a member of the C-54 family of cysteine proteases. ATG4B: Cysteine protease required for autophagy, which cleaves the C-terminal part of either MAP1LC3, GABARAPL2 or GABARAP, allowing the liberation of form I. A subpopulation of form I is subsequently converted to a smaller form (form II). Form II, with a revealed C-terminal glycine, is considered to be the phosphatidylethanolamine (PE)-conjugated form, and has the capacity for the binding to autophagosomes. Mainly expressed in the skeletal muscle, followed by brain, heart, liver and pancreas. Inhibited by N-ethylmaleimide. Belongs to the peptidase C54 family. 5 isoforms of the human protein are produced by alternative splicing.
