Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Cornifin-B (SPRR1B) ELISA Kit employs a two-site sandwich ELISA to quantitate SPRR1B in samples. An antibody specific for SPRR1B has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SPRR1B present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SPRR1B is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SPRR1B bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: All SPRR genes are closely linked within a 300-kb DNA segment on 1q21-q22.Marvin et al. (1992) characterized a cDNA clone corresponding to mRNA abundantly expressed in differentiating squamous cells originally isolated from a rabbit squamous cell library. Small Proline Rich Protein 1B is a 14-kD protein that appears to function as a component of the crosslinked envelope in squamous differentiating cells. Called cornifin, the protein has a high content of proline (31%), glutamine (20%), and cysteine (11%) and contains 13 repeats of an octapeptide (consensus sequence, EPCQPKVP) at its C terminus. In situ hybridization analysis showed that the mRNA is restricted to the suprabasal layers of the epidermis. Crosslinking of the protein appears to be mediated by transglutaminase type I.

The protein encoded by this gene is an envelope protein of keratinocytes. The encoded protein is crosslinked to membrane proteins by transglutaminase, forming an insoluble layer under the plasma membrane. Belongs to the cornifin (SPRR) family.
