3. Aspirate and add 100ul prepared Detection Reagent A. Incubate 1 hour at 37 degree C
4. Aspirate and wash 3 times
5. Add 100ul prepared Detection Reagent B. Incubate 30 minutes at 37 degree C
6. Aspirate and wash 5 times
7. Add 90ul Substrate Solution. Incubate 10-20 minutes at 37 degree C
8. Add 50ul Stop Solution. Read at 450nm immediately.
Test Principle
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Cathepsin K (CTSK). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Cathepsin K (CTSK). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Cathepsin K (CTSK), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Cathepsin K (CTSK) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
Preparation and Storage
Store at 4 degree C for 1 month/ -20 degree C for long-term (within shelf life).
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

Uniprot Description

C5aR: a family 1 G-protein coupled receptor that stimulates the GTPase activity of Gi2. Receptor for the chemotactic and inflammatory complement factor C5a. Stimulates chemotaxis, granule enzyme release and superoxide anion production. Phosphorylated in response to C5a or after stimulation of cells with phorbol esters. Expressed on monocytes, granulocytes, dendritic cells, astrocytes and microglia.

Product Notes

The Human C5aR1 c5ar1 (Catalog #AAA2709647) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA2709647 ELISA Kit recognizes Human C5aR1. AAA Biotech's Complement Component 5a Receptor 1 (C5aR1) can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). Enzyme-linked immunosorbent assay for Antigen Detection. Researchers should empirically determine the suitability of the C5aR1 c5ar1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Complement Component 5a Receptor 1 (C5aR1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
