Human Claudin 7 (CLDN7) ELISA Kit
Principle of the Assay: CLDN7 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-CLDN7 antibody and an CLDN7-HRP conjugate. The assay sample and buffer are incubated together with CLDN7-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CLDN7 concentration since CLDN7 from samples and CLDN7-HRP conjugate compete for the anti-CLDN7 antibody binding site. Since the number of sites is limited, as more sites are occupied by CLDN7 from the sample, fewer sites are left to bind CLDN7-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CLDN7 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the claudin family. Claudins are integral membrane proteins and components of tight junction strands. Tight junction strands serve as a physical barrier to prevent solutes and water from passing freely through the paracellular space between epithelial or endothelial cell sheets, and also play critical roles in maintaining cell polarity and signal transductions. Differential expression of this gene has been observed in different types of malignancies, including breast cancer, ovarian cancer, hepatocellular carcinomas, urinary tumors, prostate cancer, lung cancer, head and neck cancers, thyroid carcinomas, etc.. Alternatively spliced transcript variants encoding different isoforms have been found.[provided by RefSeq, May 2010]
