Human Chromosome associated kinesin KIF4A (KIF4A) ELISA Kit
Principle of the Assay: Ki-67 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-Ki-67 antibody and an Ki-67-HRP conjugate. The assay sample and buffer are incubated together with Ki-67-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Ki-67 concentration since Ki-67 from samples and Ki-67-HRP conjugate compete for the anti-Ki-67 antibody binding site. Since the number of sites is limited, as more sites are occupied by Ki-67 from the sample, fewer sites are left to bind Ki-67-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Ki-67 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the kinesin 4 subfamily of kinesin related proteins. The encoded protein is an ATP dependent microtubule-based motor protein that is involved in the intracellular transport of membranous organelles. This protein also associates with condensed chromosome arms and may be involved in maintaining chromosome integrity during mitosis. This protein may also be involved in the organization of the central spindle prior to cytokinesis. A pseudogene of this gene is found on chromosome X.[provided by RefSeq, Mar 2010]
