Reactivity
Human
Specificity
This assay has high sensitivity and excellent specificity for detection of CHKA. No significant cross-reactivity or interference between CHKA and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CHKA and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Competitive
Sensitivity
1.0 ng/mL
Preparation and Storage
Store all reagents at 2-8 degree C.

Intended Uses: This CHKA ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human CHKA. This ELISA kit for research use only!

Principle of the Assay: CHKA ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-CHKA antibody and an CHKA-HRP conjugate. The assay sample and buffer are incubated together with CHKA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CHKA concentration since CHKA from samples and CHKA-HRP conjugate compete for the anti-CHKA antibody binding site. Since the number of sites is limited, as more sites are occupied by CHKA from the sample, fewer sites are left to bind CHKA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CHKA concentration in each sample is interpolated from this standard curve.

NCBI Description

The major pathway for the biosynthesis of phosphatidylcholine occurs via the CDP-choline pathway. The protein encoded by this gene is the initial enzyme in the sequence and may play a regulatory role. The encoded protein also catalyzes the phosphorylation of ethanolamine. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

Uniprot Description

CHKA: Has a key role in phospholipid biosynthesis and may contribute to tumor cell growth. Catalyzes the first step in phosphatidylcholine biosynthesis. Contributes to phosphatidylethanolamine biosynthesis. Phosphorylates choline and ethanolamine. Has higher activity with choline. Belongs to the choline/ethanolamine kinase family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Human CHKA chka (Catalog #AAA7232272) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7232272 ELISA Kit recognizes Human CHKA. It is sometimes possible for the material contained within the vial of "Choline kinase Alpha (CHKA), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
