Specificity
This assay has high sensitivity and excellent specificity for detection of CDC5L. No significant cross-reactivity or interference between CDC5L and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CDC5L and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This CDC5L ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of HumanCDC5L. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: CDC5L ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CDC5L antibody and an CDC5L-HRP conjugate. The assay sample and buffer are incubated together with CDC5L-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CDC5L concentration since CDC5L from samples and CDC5L-HRP conjugate compete for the anti-CDC5L antibody binding site. Since the number of sites is limited, as more sites are occupied by CDC5L from the sample, fewer sites are left to bind CDC5L-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CDC5L concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene shares a significant similarity with Schizosaccharomyces pombe cdc5 gene product, which is a cell cycle regulator important for G2/M transition. This protein has been demonstrated to act as a positive regulator of cell cycle G2/M progression. It was also found to be an essential component of a non-snRNA spliceosome, which contains at least five additional protein factors and is required for the second catalytic step of pre-mRNA splicing. [provided by RefSeq, Jul 2008]

Uniprot Description

CDC5L: is a spliceosome protein that binds DNA and that also regulates transcription. Is a positive regulator of cell cycle G2/M progression. Is an essential component of non-snRNA spliceosomes and is required for the second catalytic step of pre-mRNA splicing. Binds adeno-pre-mRNA in an ATP-stimulated manner. Is part of a spliceosomal 'core' complex that includes CDC5L, SF2, PRLI, SPF27, HSP7C, PRPF19/SNEV, GCN1L. Identified in the spliceosome C complex. Has been associated with aneuploidy in aging cells and in tumorigenesis. The genomic region containing the gene for CDC5L (6p21.1) is frequently amplified in osteosarcomas.

Product Notes

The Human CDC5L cdc5l (Catalog #AAA7227993) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7227993 ELISA Kit recognizes Human CDC5L. It is sometimes possible for the material contained within the vial of "Cell division cycle 5 like protein (CDC5L), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
