Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of CIP2A in human serum, plasma and other biological fluids.
Principle of the Assay: The microplate provided in this kit has been pre-coated with an antibody specific to CIP2A. Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to CIP2A. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain CIP2A, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of CIP2A in the samples is then determined by comparing the O.D. of the samples to the standard curve.

CIP2A: an oncoprotein that inhibits PP2A in human malignancies. Like the SV40 small t antigen, inhibits the phospho-serine/-threonine phosphatase PP2A. Promotes c-Myc protein stability by inhibiting the dephosphorylation of phospho-serine 62. Expressed at low levels in most of nonmalignant tissues but at high levels in human malignancies including head and neck squamous cell carcinoma (HNSCC), colon cancer, and liver cancer. Antibodies against CIP2A are present in sera from many patients with gastric or prostate cancer. Two alternatively spliced human isoforms have been described.
