Intended Uses: This sandwich kit is for the accurate quantitative detection of human Calpain 7 (also known as CAPN7) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human CAPN7 antibody. CAPN7 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human CAPN7 Antibody is added and binds to CAPN7 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CAPN7 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human CAPN7. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

NCBI Description

Calpains are ubiquitous, well-conserved family of calcium-dependent, cysteine proteases. The calpain proteins are heterodimers consisting of an invariant small subunit and variable large subunits. The large subunit possesses a cysteine protease domain, and both subunits possess calcium-binding domains. Calpains have been implicated in neurodegenerative processes, as their activation can be triggered by calcium influx and oxidative stress. The function of the protein encoded by this gene is not known. An orthologue has been found in mouse but it seems to diverge from other family members. The mouse orthologue is thought to be calcium independent with protease activity. [provided by RefSeq, Jul 2008]
