Intended Uses: This BRD3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human BRD3. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: BRD3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-BRD3 antibody and an BRD3-HRP conjugate. The assay sample and buffer are incubated together with BRD3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BRD3 concentration since BRD3 from samples and BRD3-HRP conjugate compete for the anti-BRD3 antibody binding site. Since the number of sites is limited, as more sites are occupied by BRD3 from the sample, fewer sites are left to bind BRD3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BRD3 concentration in each sample is interpolated from this standard curve.

This gene was identified based on its homology to the gene encoding the RING3 protein, a serine/threonine kinase. The gene localizes to 9q34, a region which contains several major histocompatibility complex (MHC) genes. The function of the encoded protein is not known. [provided by RefSeq, Jul 2008]

BRD3: Binds hyperacetylated chromatin and plays a role in the regulation of transcription, probably by chromatin remodeling and interaction with transcription factors. Regulates transcription by promoting the binding of the transcription factor GATA1 to its targets. Regulates transcription of the CCND1 gene. A chromosomal aberration involving BRD3 is found in a rare, aggressive, and lethal carcinoma arising in midline organs of young people. Translocation t(15;9)(q14;q34) with NUT which produces a BRD3-NUT fusion protein. 2 isoforms of the human protein are produced by alternative splicing.
