Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- BMP-5 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti- BMP-5 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the BMP-5 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of BMP-5 can be calculated.

Background: Bone morphogenetic protein 5 (BMP-5) is a member of the transforming growth factor-beta superfamily of regulatory molecules. Bone morphogenetic proteins were originally identified by the ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site, and known for their ability to induce bone and cartilage development. BMP-5 is expressed in the trabecular meshwork and optic nerve head and may have a role in the development and normal function. It is also expressed in the lung and liver. It may play a role in certain cancers. Like other BMP's BMP5 is inhibited by chordin and noggin. This protein also act as an important signaling molecule within the trabecular meshwork and optic nerve head, and may play a potential role in glaucoma pathogenesis.

NCBI Description

This gene encodes a member of the bone morphogenetic protein family which is part of the transforming growth factor-beta superfamily. The superfamily includes large families of growth and differentiation factors. Bone morphogenetic proteins were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. These proteins are synthesized as prepropeptides, cleaved, and then processed into dimeric proteins. This protein may act as an important signaling molecule within the trabecular meshwork and optic nerve head, and may play a potential role in glaucoma pathogenesis. This gene is differentially regulated during the formation of various tumors. [provided by RefSeq, Jul 2008]

Product Notes

The Human BMP-5 bmp5 (Catalog #AAA355388) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA355388 ELISA Kit recognizes Human BMP-5. AAA Biotech's BMP-5 can be used in a range of immunoassay formats including, but not limited to, Quantitative sELISA (EIA). For quantitative detection of BMP-5 in human serum, plasma, body fluids, tissue lysates or cell culture supernates. Researchers should empirically determine the suitability of the BMP-5 bmp5 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "BMP-5, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
