Specificity
This assay has high sensitivity and excellent specificity for detection of SGCB. No significant cross-reactivity or interference between SGCB and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between SGCB and all the analogues, therefore, cross reaction may still exist in some cases.

Assay Type
Quantitative Competitive
Sensitivity
0.1 ng/mL

Intended Uses: This SGCB ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of HumanSGCB. This ELISA kit for research use only!

Principle of the Assay: SGCB ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-SGCB antibody and an SGCB-HRP conjugate. The assay sample and buffer are incubated together with SGCB-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SGCB concentration since SGCB from samples and SGCB-HRP conjugate compete for the anti-SGCB antibody binding site. Since the number of sites is limited, as more sites are occupied by SGCB from the sample, fewer sites are left to bind SGCB-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SGCB concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the sarcoglycan family. Sarcoglycans are transmembrane components in the dystrophin-glycoprotein complex which help stabilize the muscle fiber membranes and link the muscle cytoskeleton to the extracellular matrix. Mutations in this gene have been associated with limb-girdle muscular dystrophy.[provided by RefSeq, Oct 2008]

Uniprot Description

SGCB: Component of the sarcoglycan complex, a subcomplex of the dystrophin-glycoprotein complex which forms a link between the F-actin cytoskeleton and the extracellular matrix. Defects in SGCB are the cause of limb-girdle muscular dystrophy type 2E (LGMD2E). LGMD2E is an autosomal recessive disorder. Belongs to the sarcoglycan beta/delta/gamma/zeta family.

Protein type: Dystrophin complex; Membrane protein, integral

Chromosomal Location of Human Ortholog: 4q12

Cellular Component: dystrophin-associated glycoprotein complex; cytoskeleton; integral to plasma membrane; cytoplasm; sarcoglycan complex; sarcolemma

Biological Process: muscle development; cardiac muscle cell development; muscle fiber development

Disease: Muscular Dystrophy, Limb-girdle, Type 2e

Product Notes

The Human SGCB sgcb (Catalog #AAA7233681) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7233681 ELISA Kit recognizes Human SGCB. It is sometimes possible for the material contained within the vial of "beta sarcoglycan (SGCB), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
