Principle of the Assay: Human BCAM ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for BCAM has been precoated onto 96-well plates. Standards(Expression system for standard: NSO; Immunogen sequence: E32-A547) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for BCAM is added subsequently and then followed by washing with PBS or TBS buffer. Avidin- Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human BCAM amount of sample captured in plate.

Background/Introduction: Basal cell adhesion molecule is a protein that in humans is encoded by the BCAM gene. The BCAM gene encodes a glycoprotein expressed on cell surfaces. It is a member of the immunoglobulin superfamily of receptors and cell adhesion molecules. When expressed on erythrocytes, BCAM constitutes the Lutheran (Lu) and Auberger (Au) blood group antigens. And this protein may play a role in epithelial cell cancer and in vaso-occlusion of red blood cells in sickle cell disease. Two transcript variants encoding different isoforms have been found for this gene.

NCBI Description

This gene encodes Lutheran blood group glycoprotein, a member of the immunoglobulin superfamily and a receptor for the extracellular matrix protein, laminin. The protein contains five extracellular immunoglobulin domains, a single transmembrane domain, and a short C-terminal cytoplasmic tail. This protein may play a role in epithelial cell cancer and in vaso-occlusion of red blood cells in sickle cell disease. Polymorphisms in this gene define some of the antigens in the Lutheran system and also the Auberger system. Inactivating variants of this gene result in the recessive Lutheran null phenotype, Lu(a-b-), of the Lutheran blood group. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, May 2012]

Product Notes

The Human BCAM bcam (Catalog #AAA178575) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA178575 ELISA Kit recognizes Human BCAM. It is sometimes possible for the material contained within the vial of "BCAM, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
