Principle of the Assay: The Picokine™ Human CD80 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human CD80 with a 96-well strip plate that is pre-coated with antibody specific for CD80. The detection antibody is a biotinylated antibody specific for CD80. The capture antibody is monoclonal antibody from mouse, the detection antibody is monoclonal antibody from mouse. The kit contains recombinant Human CD80 with immunogen: Expression system for standard: NSO; Immunogen sequence: V35-N242. The kit is analytically validated with ready to use reagents. To measure Human CD80, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly propotional to Human CD80 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human CD80 in the sample.!!Background/Introduction: The protein CD80(Cluster of Differentiation 80) is a molecule found on activated B cells and monocytes which provides a costimulatory signal necessary for T cell activation and survival. It is also known as B7-1. The cDNA for B7-1 predicts a type I membrane protein, i.e., one synthesized with a signal peptide that is cleaved upon translocation across the endoplasmic membrane. The protein is predicted to contain 2 extracellular domains structurally similar to those of Ig, a hydrophobic transmembrane region, and a short cytoplasmic domain. The CD80 and CD86 genes encode B7-1 and B7-2, respectively, which are structurally similar members of the immunoglobulin superfamily expressed on a variety of hematopoietic cell types. B7-1 and B7-2 provide a costimulatory signal to T cells by interacting with CD28 and CTLA4.

The protein encoded by this gene is a membrane receptor that is activated by the binding of CD28 or CTLA-4. The activated protein induces T-cell proliferation and cytokine production.
