Principle of the Assay: MR-proANP ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-MR-proANP antibody and an MR-proANP-HRP conjugate. The assay sample and buffer are incubated together with MR-proANP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MR-proANP concentration since MR-proANP from samples and MR-proANP-HRP conjugate compete for the anti-MR-proANP antibody binding site. Since the number of sites is limited, as more sites are occupied by MR-proANP from the sample, fewer sites are left to bind MR-proANP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MR-proANP concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the membrane-spanning 4A gene family. Members of this nascent protein family are characterized by common structural features and similar intron/exon splice boundaries and display unique expression patterns among hematopoietic cells and nonlymphoid tissues. This gene encodes a B-lymphocyte surface molecule which plays a role in the development and differentiation of B-cells into plasma cells. This family member is localized to 11q12, among a cluster of family members. Alternative splicing of this gene results in two transcript variants which encode the same protein. MS4A1: This protein may be involved in the regulation of B-cell activation and proliferation. Defects in MS4A1 are the cause of immunodeficiency common variable type 5 (CVID5); also called antibody deficiency due to CD20 defect. CVID5 is a primary immunodeficiency characterized by antibody deficiency, hypogammaglobulinemia, recurrent bacterial infections and an inability to mount an antibody response to antigen. The defect results from a failure of B-cell differentiation and impaired secretion of immunoglobulins; the numbers of circulating B-cells is usually in the normal range, but can be low. Belongs to the MS4A family.
