Principle of the Assay||DDX42 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-DDX42 antibody and an DDX42-HRP conjugate. The assay sample and buffer are incubated together with DDX42-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DDX42 concentration since DDX42 from samples and DDX42-HRP conjugate compete for the anti-DDX42 antibody binding site. Since the number of sites is limited, as more sites are occupied by DDX42 from the sample, fewer sites are left to bind DDX42-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DDX42 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the Asp-Glu-Ala-Asp (DEAD) box protein family. Members of this protein family are putative RNA helicases, and are implicated in a number of cellular processes involving alteration of RNA secondary structure such as translation initiation, nuclear and mitochondrial splicing, and ribosome and spliceosome assembly. Members of this family are believed to be involved in embryogenesis, spermatogenesis, and cellular growth and division. DDX42: ATP-dependent RNA helicase. Binds to partially double- stranded RNAs (dsRNAs) in order to unwind RNA secondary structures. Unwinding is promoted in the presence of single-strand binding proteins. Mediates also RNA duplex formation thereby displacing the single-strand RNA binding protein. ATP and ADP modulate its activity: ATP binding and hydrolysis by DDX42 triggers RNA strand separation, whereas the ADP-bound form of the protein triggers annealing of complementary RNA strands. Involved in the survival of cells by interacting with TP53BP2 and thereby counteracting the apoptosis-stimulating activity of TP53BP2. Relocalizes TP53BP2 to the cytoplasm. Belongs to the DEAD box helicase family. DDX42 subfamily. 2 isoforms of the human protein are produced by alternative splicing.
