Human Arf GAP with GTPase, ANK repeat and PH domain containing protein 2 (AGAP2) ELISA Kit
Principle of the Assay: AGAP2 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-AGAP2 antibody and an AGAP2-HRP conjugate. The assay sample and buffer are incubated together with AGAP2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AGAP2 concentration since AGAP2 from samples and AGAP2-HRP conjugate compete for the anti-AGAP2 antibody binding site. Since the number of sites is limited, as more sites are occupied by AGAP2 from the sample, fewer sites are left to bind AGAP2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AGAP2 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene belongs to the centaurin gamma-like family. It mediates anti-apoptotic effects of nerve growth factor by activating nuclear phosphoinositide 3-kinase. It is overexpressed in cancer cells, and promotes cancer cell invasion. Alternatively spliced transcript variants encoding different isoforms have been described for this gene. CENTG1: a member of the centaurin family of ADP-ribosylation factor directed GTPase-activating proteins (GAPs). A GAP for ARF1 and ARF5. Two alternatively-spliced isoforms have been described. Isoform 1 is brain-specific, and isoform 2 is ubiquitously expressed, with highest levels in brain and heart. Isoform 1 participates to the prevention of neuronal apoptosis by enhancing PI3 kinase activity. It aids the coupling of metabotropic glutamate receptor 1 (GRM1) to cytoplasmic PI3 kinase by interacting with Homer scaffolding proteins, and also seems to mediate anti-apoptotic effects of NGF by activating nuclear PI3 kinase. Isoform 2 does not stimulate PI3 kinase but may protect cells from apoptosis by stimulating Akt. Regulates the adapter protein 1 (AP-1)-dependent trafficking of proteins in the endosomal system. The PH domains of isoforms 1 and 2 bind phospholipids, but they differentially regulate subcellular location. The PH domain of isoform 1 directs the protein to the nucleus, but the PH domain of isoform 2 directs it to the cytosol.
